ProductBackground:TheassaydevelopedbyEchelonBiosciencesInc.usesthefluorogenicautotaxinsubstrateFS-3.FS-3isanLPCanaloguethatisconjugatedwithbothafluorophoreandaquencher.Initsnativestatethequencherinterfereswiththefluorophore’sfluorescence.OnceautotaxincleavesFS-3,thefluorophorebecomesliberatedfromthequencher,resultinginincreasedfluorescence.
Thisassaygivesaquantitativemeasurementofautotaxinactivityinbiologicalsamplesbymeasuringchangesinfluorescenceovertime.Becausetheassayisnotendpointbased,ithastheadvantageofbeinglesssusceptIBLetoerrorarisingfrommistimingtheadditionofcomponentstodifferentsamplewells.Thefinalreadoutisinquantitativeunitsthatcorrectforinconsistenciesbetweendifferentusersequipment.
